The Effects of Superoxide Dismutase and 5, 6, 7, 8-Tetrahydrobiopterin on Hepatotoxicity Induced by Carbon Tetrachloride in Sprague-Dawley Rats

Abstract
The effects of superoxide dismutase(SOD) and 5,6,7,8 -tetrahydrobiopterin(BH4) on liver damage induced by Carbon tetrachloride(CCl4) were investigated in Sprague-Dawley rats. In this study, CCl4 was used as a chemical inducer of liver damage in rats. The levels of serum alanine aminotransferase(ALT) and aspartate aminotransferase(AST) activities, which are indices of liver cell damage , were markedly increased in CCl4-treated rats. To evaluate the effects of SOD or BH4 in the recovery of damaged hepatocytes, we co-injected these SOD or BH4 with CCl4. However, there was no perceptible decrease in the activities of ALT and AST in serum. In addition, other cell damage markers used in liver or kidney, such as bililubin, albumin, total protein, cholesterol, triglyceride, BUN and creatinine, were also tested, but the level of these markers in serum were not chanced markedly. To confirm these results obtained from the analysis of serum, histopathologic examinations were performed. In the group treated with CCl4, severe ballooning degenerations were observed. Ultrastructural features of the hepatocytes showed extreme dilatation of endoplasmic reticulum(ER). Severe ballooning degenerations and extreme dilatation of ER were also still found in the group co-treated with SOD or BH4. Nonetheless, we didn¡¯t recognize any effects of SODor BH4 in CCl4-induced liver cell damage. The level of malondialdehyde(MDA), which is represented by lipid peroxidation, was shown to be significantly higher in the CCl4-treated group compared with control. Additional treatment of SOD or BH4 in the group injected with CCl4 showed no effect in damaged liver. The enzymatic activities of SOD in the liver were also investigated, but showed no significant change. In Conclusion, the liver cell damage induced by CCl4 was not improved by simple injection of free radical scavengers such as SOD and BH4.
